Complete blood count, manual and digital cell differential
A complete blood count (CBC) was performed on a Sysmex XE 5000 Hematology Analyzer (Kobe, Japan). The Sysmex SP1000i Automatic Staining Machine / Stainer was used to slide smear preparation and peripheral blood staining
(PB) samples. At least two independent morphologists evaluated 200 cells per slide to provide a cell differential. Sysmex Cellavision DM8 produced digital cell differential after analysis of 100 cells per slide.
Immunophenotyping by flow cytometry
Immunophenotyping was performed by flow cytometry. on a Beckman Coulter FC500 five-colour flow cytometer (BC International; Nyon, Switzerland). Fluorescence tagging Antibodies for staining of samples were obtained from Beckman Coulter, EXBIO (Praha, Czech Republic), Dako. (Glostrup, Denmark) and Becton Dickinson (BD; San José, CALIFORNIA).
The samples were prepared with a whole blood lysis enumeration method and phenotypic surface characterization of PCs, as recommended. Fixation and
permeabilization was also used for the analysis of the expression of intracellular light chains.
Briefly, 50 aliquots of blood samples were incubated with the conjugated antibodies as suggested by the manufacturers and lysed with an ammonium chloride-based lysis solution (BC International; Nyon, Switzerland). For intracellular staining, 50 ll aliquots of blood samples were incubated
with the conjugated antibodies for surface staining, subsequently fixed and permeabilized with the FIX & PERM kit (Invitrogen; Carlsberg, CA), and then incubated with the Conjugated antibodies for intracellular staining.
